چکیده
Helicobacter pylori infection is chronic situation that increases the risk of development of peptic ulcer disease, mucosa-associated lymphoid tissue lymphoma (MALT), and gastric cancer. In many developing countries, such as Iran, H. pylori infect approximately 80 % of the population. Management of H. pylori infections is difficult, and relapse frequently occurs after successful eradication of the organism (Malfertheiner et al., 2007, MalekiMSc and SiavoshiPhD, 2013). Increasing rates of resistance to the antibiotic such as metronidazole and clarithromycin are result in therapy failures. Therefore, alternative treatments that include tetracyclines are recommended (Milani et al., 2012; Mansour-Ghanaei et al., 2013; Wang et al., 2014). Tetracyclines are bacteriostatic drugs which affecting the 30S subunit of the ribosome and block the binding of aminoacyl-tRNA, resulting in impaired protein biosynthesis (Gerrits et al., 2002). The tetracycline resistance to H. pylori is reported to be caused by mutations

Detection of A2142C, A2142G, and A2143G Mutations in 23s rRNA Gene Conferring Resistance to Clarithromycin among Helicobacter pylori Isolates in Kerman, Iran
Background: Clarithromycin resistance in Helicbacter pylori has been found to be associated with point mutations in 23s rRNA gene leads to reduced affinity of the antibiotic to its ribosomal target or changing the site of methylation. The aim of this study was to determine the most important point mutations in 23s rRNA gene in H. pylori that are closely related to clarith-romycin resistance amo...

OBJECTIVES To investigate the occurrence of 16S rDNA mutations associated with resistance or reduced susceptibility to tetracycline in Helicobacter pylori isolated in England and Wales, and to develop a real-time PCR assay to detect these DNA polymorphisms from culture and gastric biopsies. METHODS Tetracycline susceptibility was determined by disc diffusion. The MIC of isolates with reduced ...
متن کاملMutations in the 16S rRNA genes of Helicobacter pylori mediate resistance to tetracycline.
Low-cost and rescue treatments for Helicobacter pylori infections involve combinations of several drugs including tetracycline. Resistance to tetracycline has recently emerged in H. pylori. The 16S rRNA gene sequences of two tetracycline-resistant clinical isolates (MIC = 64 microg/ml) were determined and compared to the consensus H. pylori 16S rRNA sequence. One isolate had four nucleotide sub...

The triple-base-pair 16S rDNA mutation AGA(926-928)-->TTC mediates high-level tetracycline resistance in Helicobacter pylori. In contrast, single- and double-base-pair mutations mediated only low-level tetracycline resistance and decreased growth rates in the presence of tetracycline, explaining the preference for the TTC mutation in tetracycline-resistant H. pylori isolates.

Most Helicobacter pylori strains are susceptible to tetracycline, an antibiotic commonly used for the eradication of H. pylori. However, an increase in incidence of tetracycline resistance in H. pylori has recently been reported. Here the mechanism of tetracycline resistance of the first Dutch tetracycline-resistant (Tet(r)) H. pylori isolate (strain 181) is investigated. Twelve genes were sele...
